i have ran into this several times. Only thing fixes it is to reload or upgrade the machines firmware. FYI
The amount of pages is totally irrelevant, computers deal with data size, not pages.
A single page of text in pdf format without OCR can be as little as a few Kb, or well into the MB's in size, what matters is the content of the pages, a bunch of black text on a white background is small, the identical sized page (Letter/A4) containing a full color picture at 600DPI will be massive in size. Both are still one page.
Run a wireshark trace of the scans(small and big) and read that to find out which side is failing and why, if your printer is up to date in firmware it will have the trace feature built in

Is it possible it can only hold 20 pages in a "cache"?

I checked the setting(s) mentioned and it was already enabled. The thing that is really starting to get me is that the 20 page scan that was successful hit my inbox at roughly 1.50 MB. So the thirty page scan that failed cannot be anywhere near the 20 MB maximum. Now I'm really at a loss. We use office 365 so am I way off base in thinking the culprit might be along that path or am I totally missing it? Please advise.

How you print has no bearing on how you scan. If the file is more than 20MB it simply won't scan, if it is less than 20MB it will.
Settings wise, if a one page file works, so will 10,000 pages, there is no difference at all, but there are data limits, Xerox devices max out at 20MB, I don't know why. Especially now when most services accept 25MB.
I have tweaked the file size to the maximum with the same failed results. Question. These Xerox AltaLink C8045 MFP's all reside on a print server. Could there be any setting(s) on the print server that I should be looking at?

Assuming it is not a server side resource, go into EWS > Properties > Connectivity > Setup > SMTP (Email) > Edit
File Size Management
Change it from 10240 to 20480 > Apply
After that , if still failing, you will need to make the job less than 20MB, and to do that you will need to selectively disable things (Resolution/OCR etc) and maybe increase the compression
